When acquiring spectral data, should my single-stained controls be "as bright or brighter" than my fully-stained sample?

Yes. You may be familiar with the guideline that when acquiring compensation controls, a single-stained control should be "as bright or brighter" as compared to when that same fluorophore is used in the fully-stained sample.

When acquiring spectral data and calculating an unmixing matrix, the same approach may be considered.
