Adjust settings in the View Editor:

1. Select the Cell Segmentation Data option from the drop-down menu in the View Editor window.

2. Check the boxes for the components (channels) that you require data to be exported for (usually all components).

3. Select the desired component statistics (usually Mean and Total) to be exported.

4. Select the X and Y Position Stats. Note that these statistics are a mandatory requirement for loading data into FCS Express.

5. All other parameters are optional depending on the type of analysis being performed.

Adjust Settings in the Manual Analysis Export Settings tab

1. Specify the Export Directory destination for the exported files in the Manual Analysis > Export Settings tab.

2. Check the boxes for Component Images (multi-image TIFF) and Segmentation Maps (multi-image TIFF) in the Image export options section. Component Images and Segmentation Maps are mandatory for importing data in FCS Express. All other image views are optional.

3. Check the box for Cell Segmentation Data in the Tables to Export section.

4. Select the Use view settings radio button in the Table Fields to Export section.

5. Click Export to start the export process.

1. Open FCS Express on your computer.

2. Choose the Insert tab>2D Plots>Dot (you can also choose any plot type from the 1D Plot or 2D Plot menu).

3. Click on the FCS Express layout to create the plot.

4. Choose Perkin Elmer Vectra Files (*.tif;*.txt) from the Files of Type drop down list (image below).

5. Navigate to the folder containing your data.

6. Select the data file you would like to load into FCS Express. Please note that you may select any of the .txt or .tif files associated with the file and FCS Express will load the appropriate data (image below).

7. Click Open File.

A 2D dot plot for the data will now be displayed in FCS Express. Please see the De Novo Software website for tutorials and other resources for further working with your plots and data.

Note: Functionality for Columbus Data Base compatibility was deprecated in FCS Express 7. 

1. Select Single Cell Results from Define Results at the end of your analysis in Columbus.

2. Select the plate(s) you wish to export.

3. Designate an Export Folder.

4. Select SinglePlane TIF in the Image Data section.

5. Select Native XML in the Results section.

Note: Functionality for Columbus Data Base compatibility was deprecated in FCS Express 7. 

Importing your data for use in FCS Express could not be more simple. Before importing with the steps below please set your user options for Dialogs-->Open Data Dialog category to use the Advanced Open Data Dialog so you can easily access the Building Blocks Select a Data File tab.

Building Blocks Data

1. Open FCS Express on your computer.

2. Choose the Insert tab>2D Plots>Dot (you can also choose any plot type from the 1D Plot or 2D Plot menu).

3 Click on the FCS Express layout to create the plot.

4. Choose the Building Blocks tab in the Open Data File dialog.

5. Navigate to the folder containing your data.

6. Select the Population(s) you would like to analyze.

7. Confirm or enter the Cell Width and Cell Height.

8. Click Open.

Acapella Data

1. Open FCS Express on your computer.

2. Choose the Insert tab>2D Plots>Dot (you can also choose any plot type from the 1D Plot or 2D Plot menu).

1. Click on the FCS Express layout to create the plot.

2. Choose Select data file..

3. Choose Perkin Elmer Acapella folder from the Files of type drop down.

6. Navigate to the folder containing your data.

7. Click Open Folder
